Study Design 

Purpose:

To investigate differences between wild type MA15 infection and the deltaORF6 and ExoN mutant viruses.

Overview of Experiment:

Groups of 20 week old C57BL/6 mice will be infected.

  • MA15 at doses of 10^4 or 10^5 PFU
  • deltaORF6 at 10^5 PFU
  • ExoN at 10^4 PFU
  • mock infected. 

Mice will be euthanized on days 1, 2, 4 and 7 post-infection to measure virus load and isolate samples for measurement of virus load, lung pathology, transcriptional and proteomics analysis. 
Mock infected animals will also be harvested at each time point. 
Mice will be weighed every 24 hours to measure general disease progression and any mouse approaching 30% weight loss will be euthanized.
Due to quality of RNA, we did not run arrays on the ExoN mutant samples.
